Microcin B17 is a linear azol(in)e-containing peptide (LAP) that functions as a narrow-spectrum bactericidal peptide. The peptide targets DNA gyrase |CITS: [3086495][1846808][21904817]|. It is active mostly against Enterobacteriaceae and some Pseudomonas species, due to its limited ability to enter bacterial cells.The precursor peptide is encoded by the mcbA gene. All four |FRAME: CYS| and four of the six |FRAME: SER| residues are modified by |FRAME: EC-6.2.2.2| to azoline residues, which are further oxidized to azole rings. These modifications are catalyzed by a trimeric synthetase comprising McbB, McbC, and McbD (substrate recognition module, dehydrogenase, and YcaO proteins, respectively). Following generation of the azole rings the leader region is removed by a protease (tldD/tldE) encoded on the chromosome of E. coli. After complete processing, a two-component ABC transporter (McbE/McbF) exports the mature microcin B17 while also providing a level of self-protection.Microcin B17 is imported into target cells by two membrane proteins - the outer membrane protein OmpF and the plasma membrane protein SmbA. Once inside the cells, the peptide inhibits DNA gyrase and triggers double strand breaks in DNA.
